If say I set up a new user called ...

Bob, and set it with a password as Bob2one

Then, could i deny all access to all users on a directory. And only allow access for bob...

Logon as another user and if i try to access that directory, have a userlogin box popup to allow me to enter Bob's details?

Are you setting DENY rights in NTFS, or just removing ALLOW permissions?
